Minimize drug formulation toxicity and side-effects
Neowater, a non-toxic, biocompatible, naturally buffered vehicle offers a generic solution for minimizing drug formulation toxicity and side-effects. With Neowater one can reduce or eliminate the need for toxic excipients in the formulation. Utilizing Neowater, Do-Coop Technologies was able to formulate Cyclosporine A without any caster oils excipients that are common in the ophthalmic market, as well as in other markets thus eliminating side-effects such as irritation.

Enhance active pharmaceutical ingredient (API) bioavailability
The presence of Neowater as an excipient in a formulation has proven to alter API interaction with the cell membrane, both for hydrophilic and hydrophobic drug substances. Neowater colloidal structure and elevated amounts of carbon dioxide creates a micro-environment that has partitions of structured water (low free energy) and bulk water (high free energy). This partitioning of the water enables an increase in the kinetics of hydrophobic compounds, and decrease in hydrophilic compound kinetics, which is favorable for the drug’s in-vivo performance. A recent in-vivo study with Ketoconazole, an anti-fungal drug that was formulated in Neowater and UPW aimed to demonstrate drug performance with Neowater formulation compared to the UPW formulation. Results revealed a 300% higher Cmax value in Neowater compared to UPW formulation and a 150% increase in the AUC (area under the curve).

Enable reformulation of hydrophilic and hydrophobic compounds
Neowater’s unique post-production composition includes up to a 100-fold higher carbon dioxide concentration, neutral pH that ranges from 6-8, an enlarged surface area and buffer-like capabilities. These unique characteristics result in an enhanced ability to alter hydrophobic and hydrophilic compound bioavailability while simultaneously maintaining or improving chemical and physical stability. Recent study using Neowater as a non-toxic vehicle, Do-Coop was able to produce a liquid formulation for SG-410, a proprietary hydrophobic "brick-like" anti-cancer treatment compound that had no therapeutic window. The results revealed that the compound can be used for in-vivo studies, with at least the same activity as the DMSO based solution used for in-vitro studies. In general Improving drug efficacy while reducing its toxicity opens the way for new and extended IP, based on the extended therapeutic windows of the current APIs.

Enhanced drug stability
A variety of in-vitro results demonstrated that a Neowater-based formulation can induce stability in hydrophilic and hydrophobic drug candidates and thereby prolong their shelf life. This enhanced stability is a result of the unique structuring of the hydrogen bonds in Neowater, which reduces the free energy in the solution, eliminating the potential for chemical reactions. Furthermore, the elevated amount of carbon dioxide in Neowater serves as a buffer zone, stabilizing API in aqueous solutions. A recent study done with Meropenem, a chemically unstable carbapenem antibiotic that was hydrated in Neowater and compared to UPW, showed significantly higher stability in the Neowater solution. HPLC analysis after one week at room temperature showed a five-fold greater API concentration in Neowater compared to UPW.
